🧪 What Is It?
An autoclave is a pressure chamber that uses saturated steam at 121°C (250°F) and 15 PSI for a sustained period — typically 15 to 30 minutes — to kill bacteria, viruses, fungi, and spores through a combination of heat and moisture that denatures proteins and ruptures cell membranes. Every hospital, dental office, tattoo parlor, and microbiology lab uses one. Commercial units cost $500 to $5,000 depending on size and automation. They are all fundamentally the same thing: a pressure cooker with a gauge and a timer.
A stovetop pressure cooker reaches exactly the same temperature and pressure. The Presto 23-quart canner — the single most popular pressure cooker ever manufactured — operates at 15 PSI and 250°F. That's the textbook autoclave specification. The only differences between your pressure cooker and a medical autoclave are a printed gauge, a digital timer, and a certificate on the wall. The steam doesn't read certificates.
This is the simplest conversion in the category because there's almost nothing to convert. You're just using the pressure cooker for what it already does — generating saturated steam at sterilization temperature and pressure — and pointing it at things that aren't food. Mycologists use this method universally for sterilizing grain spawn and substrate jars. Makers use it to sterilize tools, culture media, and lab glassware. Tattoo artists in many countries are legally required to autoclave their equipment, and many use modified pressure cookers that are functionally identical to this build.

Choose the right pressure cooker. You need a stovetop model with a weighted jiggler valve, not an electric Instant Pot. Electric models max out at about 12 PSI — close but not enough for reliable sterilization. The jiggler valve is the rocking weight on top that vents excess steam when pressure exceeds the rated level. A 16-quart or larger size gives you room to sterilize multiple jars or instruments at once. Check the gasket — if it's cracked, flattened, or doesn't spring back when pressed, replace it before using.

Add a pressure gauge (optional but recommended). The jiggler valve tells you when you've hit the target pressure (it starts rocking), but it doesn't tell you the exact pressure. Drill and tap a hole in the lid or replace the jiggler with a gauge-and-valve assembly. Many pressure canner lids already have a fitting for a dial gauge — check your model. The gauge lets you verify 15 PSI precisely and monitor for pressure drops that indicate a gasket leak.

Prepare items for sterilization. Wrap instruments, tools, or glass jars in aluminum foil or place them in autoclave bags. For mycology substrate jars, fill them with grain or substrate, cover the lids with foil, and load them onto the rack. Items must not be sealed airtight — steam needs to penetrate. Leave foil loosely crimped, not sealed. Place the wire rack in the bottom of the cooker and add 2–3 inches of water below the rack. The water generates steam; the rack keeps items above the waterline.

Run the sterilization cycle. Place the loaded cooker on the stovetop over high heat with the lid locked. Wait for steam to vent steadily from the valve for 10 minutes — this purges trapped air from the chamber (air pockets prevent steam from reaching all surfaces and create cold spots). Once steam flows freely, place the jiggler weight or close the valve and allow pressure to build to 15 PSI. When the jiggler starts rocking or the gauge reads 15, reduce heat to maintain a steady gentle rock. Start the timer: 15 minutes for small items and instruments, 30 minutes for dense substrate jars, 60–90 minutes for large grain bags.

Depressurize and cool. When the timer finishes, turn off the heat and leave the cooker completely undisturbed. Do not open the valve or remove the weight. Let pressure drop to zero naturally — this takes 30–60 minutes depending on the load. Rapid depressurization causes liquids inside jars to boil over and can crack glass from thermal shock. When the gauge reads zero or the safety lock drops, open the lid tilting it away from you (residual steam is 200°F+). Remove items with heat-resistant gloves.

Verify sterilization. Check the autoclave indicator tape — it should have changed from its original color to the indicator color (usually striped). If it didn't change, the chamber didn't reach sterilization temperature, which means you have an air purge problem or a gasket leak. For mycology work, inoculate one jar as a control and leave it for a week — if it grows contaminants, your cycle wasn't long enough.
⚠️ Safety Notes
- Never open a pressurized cooker. The contents are above boiling temperature; releasing pressure instantly flash-boils all liquid in the chamber. The resulting steam explosion can cause severe burns. Always wait for pressure to drop to zero naturally.
- Inspect the gasket before every use. A worn gasket fails gradually — it holds at lower pressures but blows out at 15 PSI, spraying superheated steam from the lid joint. Replace gaskets that are stiff, cracked, or don't seat evenly.
- The safety valve exists for a reason. Never block, modify, or remove the overpressure relief valve. If the primary valve fails, the safety valve is the only thing preventing the vessel from exceeding its pressure rating.
- Once used for sterilizing non-food items (especially anything involving chemicals or biological cultures), label the cooker permanently and never return it to food use.
